FGF13 suppresses acute myeloid leukemia by regulating bone marrow niches

摘要: Fibroblast growth factor 13 (FGF13) is aberrantly expressed in multiple cancer types, suggesting its essential role in tumorigenesis. Hence, we aimed to explore its definite role in the development of acute myeloid leukemia (AML) and emphasize its associations with bone marrow niches. Results showed that FGF13 was lowly expressed in patients with AML and that its elevated expression was related to prolonged overall survival (OS). Univariate and multivariate Cox regression analyses identified FGF13 as an independent prognostic factor. A prognostic nomogram integrating FGF13 and clinicopathologic variables was constructed to predict 1-, 3-, and 5-year OS. Gene mutation and functional analyses indicated that FGF13 was not associated with AML driver mutations but was related to bone marrow niches. As for immunity, FGF13 was remarkably associated with T cell count, immune checkpoint genes, and cytokines. In addition, FGF13 overexpression substantially inhibited the growth and significantly induced the early apoptosis of AML cells. The xenograft study indicated that FGF13 overexpression prolonged the survival of recipient mice. Overall, FGF13 could serve as an independent prognostic factor for AML, and it was closely related to the bone marrow microenvironment.

摘要: Members of the fibroblast growth factor (FGF) family play pleiotropic roles in cellular and metabolic homeostasis. During evolution, the ancestor FGF expands into multiple members by acquiring divergent structural elements that enable functional divergence and specification. Heparan sulfate-binding FGFs, which play critical roles in embryonic development and adult tissue remodeling homeostasis, adapt to an autocrine/paracrine mode of action to promote cell proliferation and population growth. By contrast, FGF19, 21, and 23 coevolve through losing binding affinity for extracellular matrix heparan sulfate while acquiring affinity for transmembrane α-Klotho (KL) or β-KL as a coreceptor, thereby adapting to an endocrine mode of action to drive interorgan crosstalk that regulates a broad spectrum of metabolic homeostasis. FGF19 metabolic axis from the ileum to liver negatively controls diurnal bile acid biosynthesis. FGF21 metabolic axes play multifaceted roles in controlling the homeostasis of lipid, glucose, and energy metabolism. FGF23 axes from the bone to kidney and parathyroid regulate metabolic homeostasis of phosphate, calcium, vitamin D, and parathyroid hormone that are important for bone health and systemic mineral balance. The significant divergence in structural elements and multiple functional specifications of FGF19, 21, and 23 in cellular and organismal metabolism instead of cell proliferation and growth sufficiently necessitate a new unified and specific term for these three endocrine FGFs. Thus, the term “FGF Metabolic Axis,” which distinguishes the unique pathways and functions of endocrine FGFs from other autocrine/paracrine mitogenic FGFs, is coined.

Influence of retinoic acid on TBX1 expression in myocardial cells induced by Shh and Fgf8

摘要: The aim of this study was to explore the regulatory mechanism of retinoic acid (RA) on the TBX1 gene expression in myocardial cells. Ventricular cardiocytes were isolated from neonatal rats and cultured, and then treated with different concentrations of retinoic acid. The expression of Shh and Fgf8 at mRNA and protein levels in neonatal rat myocardial cells were measured by using RT-PCR and Western blot technique, respectively. There was basal expression of Shh and Fgf8 in the control group. When treated with 3×10 mol/L RA, we observed that the expression of Shh mRNA and protein in neonatal rat myocardial cells were up-regulated by 1.51 ( <0.05) and 1.10 times ( <0.05), respectively. In comparison with the control group, under the concentration of 5×10 mol/L RA, they were up-regulated by 2.21 ( <0.05) and 2.38 times ( <0.05) individually. Meanwhile, we could detect that the expression of Fgf8 mRNA and protein were up-regulated by 2.50 times ( <0.05) and 80% ( <0.05) separately compared with the control group after stimulation of 3×10 mol/L RA, and they were up-regulated by 3.48 ( <0.05) and 2.04 times ( <0.05) individually after stimulation of 5×10 mol/L RA. The results indicated that RA could induce the expression of Shh and Fgf8 in neonatal rat myocardial cells. At the same time, it has shown that Shh and Fgf8 were involved in the regulation process of RA on TBX1 expression.

RESPONSE OF WHEAT COMPOSITE CROSS POPULATIONS TO DISEASE AND CLIMATE VARIATION OVER 13 GENERATIONS

Since the F5 (2005), three winter wheat composite cross populations (CCPs) based on germplasm specifically suitable for low-input conditions were subjected to natural selection under organic and conventional management. In the F6, each CCP was divided into two parallel populations (12 CCPs in total) and maintained continuously until 2018. Commonly used modern cultivars with different disease susceptibilities were grown alongside to assess the agronomic performance of the CCPs. The organically managed CCPs were comparable in yield and foliar disease resistance to two continuously used reference cultivars, Achat and Capo. In contrast, under conventional management the cv. Capo outyielded the CCPs (Achat was not tested), highlighting the importance of parental cultivar choice for specific management systems. The CCPs were found to be moderately resistant to brown rust and even to the newly emerged stripe rust races prevalent in Europe since 2011. Differences between the CCPs were mainly due to parental genetic background and were significant in the first five generations, but were no longer so in the last five generations. In addition, these differences tended to vary depending on the experimental year and the environmental stresses present. In conclusion, the CCPs despite being derived from older cultivars are able to compete with more recently released reference cultivars under organic farming practices and represent a dynamic germplasm resource.

Fibroblast growth factor 21: a novel metabolic regulator from pharmacology to physiology

Fibroblast growth factor 21 (FGF21) is a member of the fibroblast growth factor family. It actually functions as endocrine hormones but does not regulate cell growth and differentiation. It is demonstrated that FGF21 acts on multiple tissue to coordinate carbohydrate and lipid metabolism, including enhancing insulin sensitivity, decreasing triglyceride concentrations, causing weight loss, ameliorating obesity-associated hyperglycemia and hyperlipidemia. Moreover, FGF21 also plays important roles in some physiological processes, such as fasting and feeding, growth hormone axis and thermogenic function of brown adipose tissue. Clinical relevance of FGF21 in humans is still unclear, and the basis and consequences of increased FGF21 in metabolic disease remain to be determined. Both the pharmacological actions and physiological roles make FGF21 attractive drug candidates for treating metabolic disease, but some questions remain to be answered. This article concentrates on recent advances in our understanding of FGF21.

Low-temperature CO oxidation over Au-doped 13X-type zeolite catalysts: preparation and catalytic activity

摘要: Au-supported 13X-type zeolite (Au/13X) was synthesized using a common deposition–precipitation (DP) method with a solution of sodium carbonate as a precipitate agent. Further testing was conducted to test for catalytic oxidation of CO. A study was conducted on the effects of different preparation conditions (i.e., chloroauric acid concentration, solution temperature, pH of solution, and calcinations temperature) on Au/13X for CO oxidation. In respect to the catalytic activity, the relationship between different the preparation conditions and gold particles in 13X zeolite was analyzed using X-ray diffraction, TEM and XPS. The activity of Au/13X catalysts in CO oxidation was dependent on the chloroauric acid concentration. From XRD results, a higher chloroauric acid concentration induced larger gold nanoparticles, which resulted in lower catalytic activity. Results revealed that higher temperatures induced higher Au loading, homogeneous deposit, and smaller gold clusters on the support of 13X, resulting in higher CO activity. Furthermore, a pH of 5 or 6 generated greater amounts of Au loading and smaller Au particles on 13X than at a pH of 8 or 9. This may be a result of an effective exchange between and Au(OH) Cl on specific surface sites of zeolite under the pH’s 5 and 6. The sample calcined at 300°C showed the highest activity, which may be due to the sample’s calcined at 200°C inability to decompose completely to metallic gold while the sample calcined at 400°C had larger particles of gold deposited on the support. It can be concluded from this study that Au/13X prepared from a gold solution with an initial chloroauric acid solution concentration of 1.5 × 10 mol·L gold solution pH of 6, solution temperature of around 90°C, and a calcination temperature of 300°C provides optimum catalytic activity for CO oxidation.

Prevalence of asthma symptoms in Golestan schoolchildren aged 6–7 and 13–14 years in Northeast Iran

Asthma is the most common chronic disease among children, and its incidences are often imminent among elementary schoolchildren. This study aimed to examine the prevalence of asthma symptoms in Golestan schoolchildren aged 6–7 and 13–14 years in Northeast Iran. The prevalence rate was compared according to age group (aged 6–7 years vs. aged 13–14 years) and gender (male vs. female). In this cross-sectional study, 1706 Iranian schoolchildren aged 6–7 and 13–14 years in Golestan Province were enrolled. Participants completed questionnaires between February and July 2014. Asthma symptoms were assessed using the questionnaire of the International Study of Asthma and Allergies in Childhood protocol in Persian. The logistic regression model was used to estimate the odds ratios (ORs) and 95% confidence intervals (CIs) of the asthma symptoms for each of the gender and age groups. The prevalence rates of “current asthma” symptoms and “asthma ever” in all the children were estimated as 9.5% and 7.5%, respectively. The prevalence of asthma (“asthma ever” and “wheezing in the past 12 months”) in junior high schoolchildren (aged 13–14 years) is higher than that in elementary schoolchildren (aged 6–7 years) (<0.05). The prevalence of the severity of wheezing in girls is lower than that in boys (OR= 1.7, 95%CI= 1.06–2.96, = 0.02). Asthma is still a major public health problem. This study shows that the prevalence of the asthma symptoms in boys is lower than that in girls in both age groups, and the severity of asthma in girls is higher than that in boys aged 13–14 years.

Separating method and dynamic processes of Nano-Al13

摘要: In order to investigate the characteristics of pure Nano-Al, Nano-Al was separated and purified from a series of poly-aluminum chloride (PAC) solutions which had the same Al percentage but different total Al concentrations, by using column chromatography, ethanol-acetone resolving and SO/Ba displacement. The Al species yield was characterized by Al-ferron timed complexation spectrophotometry and Al-NMR (nuclear magnetic resonance). The coagulation efficiency of Nano-Al, PAC and AlCl in synthetic water was also investigated by Jar tests. The dynamic process and aggregation state of kaolin suspensions coagulating with Nano-Al, PAC and AlCl were similarly investigated using a photometric dispersion analyzer 2000 (PDA2000). The experimental results indicated that the ethanol-acetone resolving method was simple and could separate the PAC solution at different concentrations, while column chromatography could separate PAC solutions at low concentrations. The SO/Ba displacement method could separate PAC solutions at high concentrations. However, extra inorganic cation and anion could be added in the solution during separation. The coagulation efficiency and dynamic experimental results showed that Nano-Al with high positive-charged species was effective in removing turbidity and color. The dynamic process results showed that Nano-Al also had the best recovery capability after shearing compared with PAC and AlCl because the Nano-Al conformation is more effective in charge neutralization.

A novel method for reconstruction in laparoscopic pancreaticoduodenectomy: an experience of 13 cases

摘要: Laparoscopic pancreaticoduodenectomy (LPD) is a challenging operation to general surgeon. Up to date, only about 135 cases have been reported, 16 cases in China, 119 cases outside China. The reconstruction of alimentary system is a key procedure to ensure success of the whole surgery. It is worth investigating the methods of reconstruction in LPD. A retrospective study is made to investigate the methods of reconstruction in LPD. We analyze 13 cases of LPD performed in our center. Child s or modified Child s method was used to make the reconstruction in our practice. We tried three methods to make the anastomosis of pancreaticojejunostomy, including end-to-end dunking binding pancreaticojejunostomy in two cases, end-to-end dunking pancreaticojejunostomy using interrupted suture in two cases, and duct-to-jejunal end-to-side embedding pancreaticojejunostomy in nine cases. The clinical data was collected and analyzed. Three of four patients, who underwent end-to-end pancreaticojejunostomy, had a little pancreatic leakage, especially in the first case. None of other nine patients, who underwent duct-to-jejunal end-to-side embedding pancreaticojejunostomy, was detected to have pancreatic leakage, and the operating time of these nine cases was less than other four cases. Duct-to-jejunal end-to-side embedding pancreaticojejunostomy is a safe and efficient method of reconstruction in LPD.

“十三五”期间乃至未来一段时间内,我国养殖产品需求仍呈刚性增长,但是养殖业面临资源短缺日趋严重、环境制约日益加剧等不利局面,同时面临国际竞争对国内产业冲击加剧等新的形势,未来我国养殖业必须走可持续发展道路。本文基于对我国养殖业的发展现状、可持续发展所面临的挑战,形成对我国养殖业可持续发展形势的基本判断,针对“十三五”,凝练了养殖业可持续发展的战略目标、基本原则和战略重点,并提出了“进一步明确养殖业在现代农业中的战略主导地位”“合理利用国际市场保障我国食物安全”“加快启动和实施一批养殖科技重点研发计划”等重大建议,为我国养殖业加快发展方式转变、实现现代化提供理论支撑。

“十三五”生态文明建设的目标与重点任务

摘要:

十三五”时期是我国加快推进生态文明建设、全面建成小康社会的关键时期。基于对生态文明建设面临的八大重要挑战的总体判断,提出了“十三五”时期生态文明建设目标和指标建议,提出了生态文明建设的九大重点任务,包括实施绿色拉动战略驱动产业转型升级、提高资源能源效率建设节约型社会、以重大工程带动生态系统量质双升、着力解决危害公众健康的突出环境问题、划定并严守生态保护红线体系、推进新型城镇化战略统筹城乡发展、开展国家生态资产家底清查核算与监控评估平台建设、全面开展全民生态文明新文化活动、实施生态文明工程科技支撑重大专项;最后提出了五项保障的条件与政策建议。

Rdh13 deficiency weakens carbon tetrachloride-induced liver injury by regulating Spot14 and Cyp2e1 expression

摘要: Mitochondrion-localized retinol dehydrogenase 13 (Rdh13) is a short-chain dehydrogenase/reductase involved in vitamin A metabolism in both humans and mice. We previously generated knockout mice and showed that Rdh13 deficiency causes severe acute retinal light damage. In this study, considering that Rdh13 is highly expressed in mouse liver, we further evaluated the potential effect of Rdh13 on liver injury induced by carbon tetrachloride (CCl ). Although Rdh13 deficiency showed no significant effect on liver histology and physiological functions under regular culture, the mice displayed an attenuated response to CCl -induced liver injury. Their livers also exhibited less histological changes and contained lower levels of liver-related metabolism enzymes compared with the livers of wild-type (WT) mice. Furthermore, the mice had Rdh13 deficiency and thus their liver cells were protected from apoptosis, and the quantity of their proliferative cells became lower than that in WT after CCl exposure. The ablation of gene decreased the expression levels of thyroid hormone-inducible nuclear protein 14 (Spot14) and cytochrome P450 (Cyp2e1) in the liver, especially after CCl treatment for 48 h. These data suggested that the alleviated liver damage induced by CCl in mice was caused by Cyp2e1 enzymes, which promoted reductive CCl metabolism by altering the status of thyroxine metabolism. This result further implicated Rdh13 as a potential drug target in preventing chemically induced liver injury.

摘要: This paper highlights the use of situated artificial institution (SAI) within a hybrid, interactive,normative multi-agent system to regulate human collaboration in crisis management. Norms regulate the actions of human actors based on the dynamics of the environment in which they are situated. This dynamics results from both environment evolution and actors’ actions. Our objective is to situate norms in the environment in order to provide a context-aware crisis regulation. However, this coupling must be a loose one to keep both levels independent and easyto-change in order to face the complex and changing crisis situations. To that aim, we introduce a constitutive level between environmental and normative states providing a loose coupling of normative regulation with environment evolution. Norms are thus no more referring to environmental facts but to status functions, i.e., the institutional interpretation of environmental facts through constitutive rules. We present how this declarative and distinct SAI modelling succeeds in managing the crisis with a context-aware crisis regulation.
